**Lirilumab + ipilimumab** is an investigational combination of two immune checkpoint inhibitor monoclonal antibodies, developed for oncology indications. **Lirilumab** is a fully human monoclonal antibody that targets inhibitory killer cell immunoglobulin-like receptors (KIRs) expressed predominantly on natural killer (NK) cells and some T cells, potentiating innate immunity by blocking inhibitory signals to NK cells. **Ipilimumab** is a monoclonal antibody that binds to cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4), a negative regulator of T cell activation, thereby potentiating adaptive immune responses. The combination was evaluated in Phase 1 studies for advanced or refractory solid tumors based on the hypothesis that simultaneous modulation of innate and adaptive immunity may provide enhanced antitumor activity compared to either agent alone. This combination is not approved and development is limited to early phase trials[1][2][3][5].
