Overview

Lisdexamfetamine is primarily used for attention deficit hyperactivity disorder (ADHD) and binge eating disorder. Antidepressants commonly co-prescribed include selective serotonin reuptake inhibitors (SSRIs), serotonin-norepinephrine reuptake inhibitors (SNRIs), tricyclic antidepressants, or atypical agents such as bupropion. The mechanism of action for lisdexamfetamine involves increasing synaptic concentrations of dopamine and norepinephrine by promoting their release and inhibiting reuptake after conversion to dextroamphetamine. Antidepressants act via various mechanisms depending on class—most commonly by modulating serotonergic and/or noradrenergic neurotransmission. Combination therapy may be used in patients with comorbid ADHD and mood disorders or in cases where augmentation is needed for residual depressive symptoms. Clinical studies have shown that adding lisdexamfetamine to ongoing antidepressant therapy can improve executive dysfunction and depressive symptoms in adults with major depressive disorder[4]. However, this combination increases the risk of drug interactions—especially serotonin syndrome when combined with serotonergic antidepressants—and may increase cardiovascular side effects such as hypertension or tachycardia[2][3][6][7].
