Overview

Livmoniplimab + budigalimab is an investigational combination of two monoclonal antibodies under development for advanced solid tumors, especially hepatocellular carcinoma (HCC) and non-small cell lung cancer (NSCLC). Livmoniplimab (ABBV-151) is a first-in-class antibody targeting the glycoprotein-A repetitions predominant (GARP)–TGF-β1 complex, thereby preventing the release of active TGF-β1 and reducing immunosuppressive signaling in the tumor microenvironment. Budigalimab (ABBV-181) is an anti-PD-1 monoclonal antibody that blocks the PD-1 immune checkpoint, enhancing T-cell–mediated antitumor activity. The rationale for the combination is to reverse TGF-β1–mediated immunosuppression while providing immune checkpoint blockade, thereby improving antitumor immune responses. Clinical trials are ongoing in advanced/metastatic HCC and NSCLC, with additional research in other advanced solid tumors[2][3][4][5][6][7][8].
