LJM716 + cetuximab is an investigational combination therapy composed of LJM716, a fully human monoclonal antibody targeting human epidermal growth factor receptor 3 (HER3), and cetuximab, a chimeric monoclonal antibody that targets epidermal growth factor receptor (EGFR/HER1). LJM716 binds to HER3 and locks it into an inactive conformation, inhibiting both ligand-induced and ligand-independent HER3 activation, thus interrupting downstream signaling crucial for cancer cell survival and drug resistance[1][2]. Cetuximab binds to EGFR, blocking ligand binding and receptor activation, and has demonstrated antitumor activities in squamous cell carcinoma and colorectal cancer. The rationale for combining these agents is based on the frequent co-activation of HER3 and EGFR in various cancers, which can render single-agent therapies less effective and contribute to acquired resistance[1]. Preclinical evidence and early clinical studies suggest synergistic benefits of dual HER3/EGFR blockade, particularly in head and neck and esophageal squamous cell carcinomas[1].
