LJM716 + trastuzumab is a combination therapy consisting of two monoclonal antibodies targeting different components of the HER signaling pathway. LJM716 (elgemtumab) is a fully human anti-HER3 IgG1 monoclonal antibody that binds a conformational epitope that traps HER3 in the inactive conformation, preventing both ligand-induced and ligand-independent activation of HER3. Trastuzumab is a well-established HER2-targeted monoclonal antibody. This combination aims to overcome resistance mechanisms in HER2-positive cancers by simultaneously inhibiting HER2 and HER3 signaling pathways.
