Drug intelligence / Profile preview
LK101 + tislelizumab + pemetrexed is an investigational combination regimen being evaluated as a first-line maintenance therapy for patients with locally advanced or metastatic non-small cell lung cancer (NSCLC). The regimen consists of LK101, a personalized neoantigen mRNA-transfected dendritic cell (DC) cancer vaccine developed by Likang Life Sciences; tislelizumab, a humanized IgG4 monoclonal antibody targeting programmed cell death protein 1 (PD-1) developed by BeiGene; and pemetrexed, a folate analog metabolic inhibitor chemotherapy. LK101 is custom-manufactured for each patient by transfecting autologous dendritic cells with mRNA encoding up to 30 patient-specific tumor neoantigens to prime neoantigen-specific T-cell responses. Tislelizumab blocks PD-1 to relieve T-cell inhibition, while pemetrexed inhibits folate-dependent enzymes involved in DNA synthesis. In a Phase 1b/2a investigator-initiated trial (NCT05886439), the combination demonstrated manageable safety and preliminary antitumor activity as first-line maintenance therapy in patients with advanced NSCLC without actionable driver alterations and with PD-L1 TPS ≥1%.
