LM-108 + anti-PD-1 antibody is a combination immunotherapy regimen consisting of LM-108 (cafelkibart), a Fc-optimized anti-CCR8 monoclonal antibody, and an anti-PD-1 antibody (such as pembrolizumab or toripalimab). LM-108 selectively depletes tumor-infiltrating regulatory T cells (Tregs) via antibody-dependent cell-mediated cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP), targeting CCR8 to overcome immune resistance within the tumor microenvironment. When combined with an anti-PD-1 antibody, which blocks PD-1-mediated immune checkpoint inhibition on T cells, this regimen aims to both remove immunosuppressive Tregs and enhance T-cell activation, thereby restoring and amplifying antitumor immune responses. The combination has shown promising clinical activity and a manageable safety profile in patients with refractory or resistant solid tumors, particularly gastric and pancreatic cancers[1][2][3].
