LM-302 + cadonilimab + capecitabine is a combination therapy consisting of three components: 1. **LM-302**: A novel antibody-drug conjugate (ADC) targeting Claudin 18.2 (CLDN18.2), which is highly expressed in gastric/gastroesophageal junction (GEJ), pancreatic, and biliary tract cancers. LM-302 consists of a CLDN18.2-specific antibody linked to the cytotoxic agent monomethyl auristatin E (MMAE) through a cleavable VC-PAB linker. 2. **Cadonilimab**: A tetravalent bispecific antibody targeting both PD-1 and CTLA-4 immune checkpoints. It features a unique design with crystalline fragments and mutations that eliminate Fc receptor and complement-mediated cytotoxic effects, potentially leading to better safety profiles compared to traditional checkpoint inhibitor combinations. 3. **Capecitabine**: An oral fluoropyrimidine chemotherapy drug that is converted to 5-fluorouracil in the body. This combination approach aims to leverage multiple mechanisms of action: targeted cytotoxicity against CLDN18.2-expressing tumor cells (LM-302), dual immune checkpoint inhibition to enhance T-cell responses (cadonilimab), and traditional chemotherapy (capecitabine).
