LM-302 + capecitabine is an investigational combination therapy composed of LM-302, an antibody-drug conjugate (ADC) targeting Claudin 18.2 (CLDN18.2), and capecitabine, a well-established oral prodrug of 5-fluorouracil (5-FU). LM-302 consists of a claudin 18.2-specific antibody linked to the cytotoxic payload monomethyl auristatin E (MMAE) via a cleavable valine-citrulline-p-aminobenzylcarbamate (VC-PAB) linker and is designed to deliver targeted cytotoxicity to CLDN18.2-expressing gastric, gastroesophageal, pancreatic, and biliary tract cancer cells through endocytosis and intracellular payload release. Capecitabine acts as a cytotoxic antimetabolite that interferes with DNA synthesis in rapidly dividing cancer cells. The combination is under clinical investigation for its potential to enhance anti-tumor efficacy in CLDN18.2-positive gastrointestinal cancers, particularly in advanced or refractory settings[1][3][7].
