Overview
LM302 + toripalimab is an investigational combination therapy for advanced solid tumors, particularly CLDN18.2-positive gastrointestinal cancers such as gastric, gastroesophageal junction, and pancreatic cancer. LM302 (also known as TPX-4589 or BMS-986476) is an antibody-drug conjugate targeting Claudin18.2, a protein highly expressed in several gastrointestinal malignancies. It delivers a cytotoxic payload directly to tumor cells expressing Claudin18.2, thereby inhibiting their proliferation[6][8]. Toripalimab is a monoclonal antibody that inhibits the programmed death receptor 1 (PD‑1), blocking its interaction with PD-L1 and PD-L2 to enhance immune-mediated tumor cell killing[1]. The combination aims to leverage both direct cytotoxicity against tumor cells and immune checkpoint inhibition for synergistic antitumor activity[3][5].
