LMP2-loaded dendritic cell vaccine + DUK-CPG-001 is an investigational combination immunotherapy developed at Duke University for the treatment of Epstein-Barr Virus (EBV)-positive Hodgkin and non-Hodgkin lymphomas. The regimen consists of an autologous dendritic cell vaccine pulsed with the EBV Latent Membrane Protein 2 (LMP2) antigen, which is designed to stimulate a specific cytotoxic T-lymphocyte response against tumor cells expressing EBV antigens. This is co-administered with DUK-CPG-001, a synthetic oligonucleotide that acts as a Toll-like receptor 9 (TLR9) agonist. DUK-CPG-001 functions as an adjuvant, enhancing the maturation and activity of the dendritic cells and promoting a robust Th1-type immune response. The therapy is typically evaluated in the setting of autologous stem cell transplant to consolidate remission and prevent relapse.
