This investigational combination therapy, developed by Baylor College of Medicine, consists of autologous LMP2a-specific cytotoxic T-lymphocytes (CTLs) administered following lymphodepletion with a CD45 monoclonal antibody. The therapy is designed to treat patients with relapsed Epstein-Barr virus (EBV)-positive Hodgkin's lymphoma or non-Hodgkin's lymphoma. The CTLs are T cells expanded ex vivo to specifically target the EBV latent membrane protein 2A (LMP2a), an antigen expressed by malignant cells in EBV-associated lymphomas. The pre-conditioning step with CD45 antibodies is intended to deplete the patient's existing immune cells, thereby creating a favorable environment for the in vivo expansion and persistence of the adoptively transferred T cells. This approach aims to overcome the immune evasion mechanisms typically employed by EBV-infected tumor cells.
