LN-145 + pembrolizumab is an investigational combination therapy developed by Iovance Biotherapeutics. It combines LN-145, an autologous tumor-infiltrating lymphocyte (TIL) cell therapy, with pembrolizumab, a PD-1-blocking monoclonal antibody. LN-145 consists of patient-derived TILs harvested from tumor tissue, expanded ex vivo, and reinfused following nonmyeloablative lymphodepletion. The combination aims to enhance the anti-tumor immune response by providing a large population of tumor-reactive T cells (LN-145) while simultaneously blocking the PD-1/PD-L1 inhibitory pathway (pembrolizumab) that often limits T-cell activity in the tumor microenvironment. This regimen is primarily being evaluated in Phase 2 trials for solid tumors, including non-small cell lung cancer (NSCLC), head and neck squamous cell carcinoma (HNSCC), and cervical cancer, particularly in patients who have not previously received immune checkpoint inhibitors.
