Overview

LN-4305 and LN-4311 are next-generation antibody-drug conjugates (ADCs) targeting Nectin-4, a cell surface protein highly expressed in various solid tumors. Developed through a collaboration between Nectin Therapeutics and LigaChem Biosciences using the ConjuAll™ technology, these assets utilize a humanized monoclonal antibody conjugated to different payloads. LN-4305 features a β-glucuronide cleavable linker with an exatecan (topoisomerase I inhibitor) payload, while LN-4311 utilizes a monomethyl auristatin E (MMAE) payload. Preclinical studies indicate that both ADCs exhibit superior potency and a wider therapeutic index compared to enfortumab vedotin, demonstrating efficacy in models of triple-negative breast cancer (TNBC), lung cancer, and ovarian cancer, including those resistant to enfortumab vedotin.
