Lobaplatin + pemetrexed is a combination chemotherapy regimen consisting of lobaplatin, a third-generation platinum-based antineoplastic agent, and pemetrexed, a multi-targeted antifolate chemotherapeutic. Lobaplatin exerts its cytotoxic effect primarily through DNA crosslinking, leading to inhibition of DNA replication and transcription. Pemetrexed inhibits several folate-dependent enzymes involved in the synthesis of thymidine and purine nucleotides (notably thymidylate synthase), thereby disrupting DNA and RNA synthesis. This combination has been investigated for use in patients with metastatic breast cancer who have failed prior anthracycline or taxane therapy[1]. The regimen is administered intravenously every 21 days.
