Lobaplatin + raltitrexed is an investigational combination chemotherapy regimen composed of two small molecule agents, lobaplatin and raltitrexed. Lobaplatin is a third-generation platinum-based antineoplastic agent that forms DNA crosslinks, thereby inhibiting DNA replication and transcription, leading to apoptosis in cancer cells. Raltitrexed is a quinazoline antifolate that selectively inhibits thymidylate synthase, blocking the synthesis of thymidine required for DNA replication and repair. This combination has been explored primarily in the treatment of solid tumors such as hepatocellular carcinoma (HCC) and colorectal cancer, often as part of transarterial chemoembolization (TACE) or drug-eluting bead TACE (DEB-TACE) regimens for unresectable or recurrent disease[2]. The rationale for combining these agents lies in their complementary mechanisms targeting different aspects of tumor cell proliferation.
