Lomustine + procarbazine is a combination of two oral chemotherapy agents used primarily in the treatment of high-grade gliomas, including anaplastic astrocytoma, oligodendroglioma, and oligoastrocytoma. Lomustine is a nitrosourea alkylating agent that acts by causing DNA and RNA cross-linking and alkylation, leading to cytotoxicity through inhibition of DNA synthesis. Procarbazine is a hydrazine derivative with cell cycle phase-nonspecific activity; it inhibits protein, RNA, and DNA synthesis via free-radical damage to DNA and also has monoamine oxidase inhibiting properties. This regimen may be used as adjuvant therapy or for recurrent disease when other regimens are not suitable[5][6][8].
