Overview

This is a combination of two drugs with a significant interaction. Lomustine (CCNU) is a lipophilic nitrosourea anticancer agent that undergoes hydrolysis to form reactive metabolites that alkylate and cross-link DNA. Valproic acid is an anticonvulsant that enhances GABAergic activity. When combined, valproic acid may impair lomustine metabolism and increase its toxicity, while lomustine may increase the toxic effects of valproic acid.
