Loncastuximab tesirine + durvalumab is an investigational combination therapy being evaluated for the treatment of relapsed or refractory B-cell lymphomas, including diffuse large B-cell lymphoma (DLBCL), mantle cell lymphoma (MCL), and follicular lymphoma (FL). Loncastuximab tesirine is an antibody-drug conjugate targeting CD19 on B cells; it consists of a humanized monoclonal antibody linked to a pyrrolobenzodiazepine (PBD) dimer cytotoxin. Upon binding to CD19, the conjugate is internalized and releases the PBD toxin, which induces DNA interstrand cross-links leading to cell death. Durvalumab is a monoclonal antibody that inhibits programmed death-ligand 1 (PD-L1), thereby enhancing anti-tumor immune responses.
