**LOOP33 + 123 CAR-T** is a dual-target, bispecific chimeric antigen receptor T cell therapy that is engineered to recognize both **CD33** and **CD123** antigens, primarily for the treatment of **acute myeloid leukemia (AML)**. Developed via lentiviral transduction of T cells from healthy donors, the therapy uses a looped configuration of CAR sequences, which enhances the ability of the CAR-T cells to efficiently eliminate AML cells even when tumor cells downregulate or lose either CD33 or CD123. Compared to monovalent (single-antigen) CAR-T therapies, Loop33 + 123 CAR-T demonstrates superior anti-leukemic activity, efficiency in preclinical immune escape models, and prolongs survival in animal models with minimal observed toxicity.
