This drug is a combination of three antiretroviral agents used for the treatment of HIV-1 infection in adults. **Lopinavir** is a protease inhibitor (PI) that prevents HIV protease from cleaving viral polyproteins, thereby disrupting viral maturation. **Ritonavir** is itself a PI, but is mainly used here as a pharmacokinetic booster, inhibiting CYP3A to significantly increase lopinavir plasma levels. **Efavirenz** is a non-nucleoside reverse transcriptase inhibitor (NNRTI), which binds directly to reverse transcriptase and blocks the RNA-dependent DNA polymerase activity of HIV-1, preventing viral replication. This triple combination is considered a nucleoside reverse transcriptase inhibitor (NRTI)-sparing regimen and may be used in patients intolerant to NRTIs or at risk of NRTI-associated toxicities. Clinical studies have shown high virologic success rates and sustained immunologic response for up to 48 weeks, though increased toxicity compared to standard NRTI-containing regimens has been noted[1][2][4][5].
