This is a combination of three antiviral drugs: lopinavir, ritonavir, and remdesivir. Lopinavir and ritonavir are both HIV protease inhibitors; they act by inhibiting viral proteases essential for the maturation of infectious viral particles. Ritonavir also inhibits cytochrome P450 3A4, boosting the plasma levels of lopinavir. Remdesivir is a nucleotide analog prodrug that inhibits viral RNA-dependent RNA polymerase, blocking viral RNA synthesis through delayed chain termination after incorporation into nascent viral RNA. This triple combination has been investigated for enhanced antiviral efficacy against SARS-CoV-2 in COVID-19 due to their complementary mechanisms—protease inhibition and polymerase inhibition[1][4][8].
