Lorcaserin + phentermine placebo refers to a specific treatment arm used in clinical safety and tolerability trials, most notably the 12-week pilot study NCT01987427 sponsored by Eisai Inc. This arm consists of active lorcaserin hydrochloride (10 mg twice daily) combined with a placebo matching the appearance and dosing schedule of phentermine. Lorcaserin is a small molecule selective serotonin 2C (5-HT2C) receptor agonist that promotes satiety by activating receptors on anorexigenic pro-opiomelanocortin neurons in the hypothalamus. The combination was evaluated to compare the safety profile of lorcaserin monotherapy against its combination with phentermine in overweight and obese adults. Although lorcaserin was previously FDA-approved for chronic weight management, it was withdrawn from the market in 2020 due to safety concerns regarding an increased risk of cancer.
