Lorigerlimab + docetaxel + prednisone is a three-drug combination under investigation for the treatment of metastatic castration-resistant prostate cancer (mCRPC). Lorigerlimab is an investigational bispecific IgG4 DART (Dual-Affinity Re-Targeting) molecule engineered to concurrently target and block both programmed cell death protein 1 (PD-1) and cytotoxic T-lymphocyte-associated protein 4 (CTLA-4), potentially enhancing immune-mediated antitumor responses with preferential activity within the tumor microenvironment. Docetaxel is a microtubule inhibitor chemotherapeutic agent that disrupts cell division by stabilizing microtubules, leading to apoptosis in rapidly dividing tumor cells. Prednisone is a synthetic glucocorticoid, used primarily to manage chemotherapy-associated side effects, and as a standard component of mCRPC regimens. The combination is currently being evaluated for its efficacy and safety in phase 2 trials as a first-line regimen for mCRPC[3][4][5].
