Lotemax Gel + Restasis is a combination therapeutic regimen evaluated in clinical trials (notably NCT02028312) for the treatment of dry eye disease. This regimen utilizes Lotemax Gel (loteprednol etabonate ophthalmic gel 0.5%), a topical corticosteroid, as an induction therapy to provide rapid anti-inflammatory relief and improve the tolerability of Restasis (cyclosporine ophthalmic emulsion 0.05%). Loteprednol etabonate acts as an agonist at glucocorticoid receptors to suppress ocular inflammation, while cyclosporine is a calcineurin inhibitor immunosuppressant that increases tear production by inhibiting T-cell activation. The combination is designed to mitigate the stinging sensation often associated with the initiation of cyclosporine therapy and to accelerate the improvement of clinical signs and symptoms of dry eye.
