Drug intelligence / Profile preview
**LOXL2 inhibitor (podocyte-specific)** refers to a therapeutic strategy or class of investigational agents designed to selectively inhibit Lysyl Oxidase-Like 2 (LOXL2) within renal podocytes. LOXL2 is an enzyme responsible for catalyzing the cross-linking of collagen and elastin in the extracellular matrix, and its aberrant upregulation is strongly associated with tissue fibrosis and remodeling. In the kidney, podocyte-specific LOXL2 expression has been shown to drive sterile inflammation, NLRP3 inflammasome priming, and downstream pro-fibrotic TGF-β/Smad signaling under metabolic and inflammatory stress. Preclinical research, including genetic knockout mouse models (such as *Nphs2-Cre:Loxl2-/-*) and CRISPR/Cas9 gene silencing, has demonstrated that targeting podocyte LOXL2 significantly reduces albuminuria, preserves podocyte structural integrity, and attenuates kidney injury in models of diabetic kidney disease (DKD) and acute kidney injury (AKI) on chronic kidney disease (CKD). Currently, this represents an active area of target validation rather than a single designated clinical-stage drug candidate.
