Overview

UI116 is a fixed-dose combination (FDC) small molecule drug candidate developed by Korea United Pharm as part of its incrementally modified drug (IMD) pipeline. It combines loxoprofen, a non-steroidal anti-inflammatory drug (NSAID) of the propionic acid class, with rebamipide, a gastroprotective agent. Loxoprofen acts as a prodrug that is metabolized into an active form which non-selectively inhibits cyclooxygenase-1 (COX-1) and cyclooxygenase-2 (COX-2) enzymes, thereby reducing the synthesis of prostaglandins involved in pain and inflammation. Rebamipide is included to mitigate the gastrointestinal side effects typically associated with NSAID therapy; it works by stimulating the production of endogenous prostaglandins in the gastric mucosa, increasing mucosal blood flow, and scavenging reactive oxygen species. This combination is intended to provide effective analgesia for conditions such as rheumatoid arthritis and osteoarthritis while improving patient safety and compliance by reducing the risk of gastric mucosal injury.
