LP11 + OVA8 + CpG + LLP2A is a programmable peptide nucleic acid (PNA)-based nanovaccine (PVN) designed for anticancer immune activation. The platform utilizes an 11-mer PNA scaffold (LP11) to facilitate the "one-pot" assembly of a PNA-modified ovalbumin antigenic peptide (OVA8/SIINFEKL), a CpG adjuvant, and a PNA-derivatized LLP2A ligand. The LLP2A ligand specifically targets the activated α4β1 integrin (VLA-4) expressed on both immune cells (such as dendritic cells) and melanoma cells. By co-delivering the antigen and the Toll-like receptor 9 (TLR9) agonist CpG, the nanovaccine enhances antigen presentation and elicits robust CD8+ T cell and natural killer (NK) cell responses, leading to significant tumor regression in syngeneic melanoma models.
