LTX-315 + pembrolizumab is a combination therapy under clinical investigation for the treatment of advanced solid tumors, including triple-negative breast cancer and melanoma. LTX-315 is a synthetic nonameric cationic oncolytic peptide derived from bovine lactotransferrin, designed to induce immunogenic cell death in tumor cells through membranolytic activity. This process leads to the release of damage-associated molecular patterns (DAMPs) such as calreticulin, ATP, HMGB1, and type I interferons, which promote robust local and systemic antitumor immune responses by enhancing T-cell infiltration into the tumor microenvironment[1][5][6]. Pembrolizumab is a monoclonal antibody that inhibits programmed cell death protein 1 (PD-1), thereby blocking an immune checkpoint pathway and enabling T-cell-mediated antitumor activity. The combination aims to leverage LTX-315’s ability to prime tumors for immune attack with pembrolizumab’s checkpoint inhibition to enhance overall therapeutic efficacy[6]. Clinical studies have shown that this combination can generate de novo T-cell responses and has demonstrated anti-tumor activity in patients with advanced solid malignancies[6].
