LTX-315 + tumor-infiltrating lymphocytes is a combination approach involving the oncolytic peptide LTX-315 and the adoptive transfer of tumor-infiltrating lymphocytes (TILs). LTX-315 is a synthetic, short oncolytic peptide designed to disrupt tumor cell membranes and induce immunogenic cell death, resulting in rapid necrosis of injected tumors, release of tumor antigens, and conversion of "cold" (immunologically inactive) tumors into "hot" (T-cell-infiltrated) tumors. The peptide promotes a pro-inflammatory tumor microenvironment and robust infiltration of cytotoxic CD8+ T cells. TIL therapy involves expanding tumor-infiltrating lymphocytes isolated from a patient’s tumor ex vivo and re-infusing them into the patient to target cancer cells. While both components individually act via immunomodulation, the rationale for their combination is to enhance tumor destruction and immune priming: LTX-315 increases tumor antigen exposure and T-cell recruitment, thereby potentially augmenting the efficacy of adoptively transferred TILs in eliminating residual or disseminated cancer cells[3][5][7].
