**LU AF20513 + booster vaccine** is a combination immunotherapy regimen intended to treat **Alzheimer’s disease**. LU AF20513 is an active immunization vaccine composed of a synthetic peptide containing repeats of the first 12 amino acids of **amyloid β (Aβ)** interspersed with helper T-cell (Th) epitopes (P2 and P30) derived from tetanus toxoid. The **booster vaccine** (administered prior to the main vaccine) is intended to activate preexisting memory T cells from previous tetanus vaccinations, leading to improved T helper cell responses, greater B cell stimulation, and increased antibody production against monomeric Aβ. The approach aims to clear amyloid plaques while avoiding autoimmune T-cell responses and is specifically optimized for older adults with diminished immunocompetence. Preclinical studies showed the vaccine induces high anti-Aβ antibody titers, reduces amyloid pathology, and is neuroprotective.
