This is a combination of **lucatumumab**, a fully human monoclonal antibody targeting CD40, and **bendamustine**, a bifunctional alkylating agent. Lucatumumab acts by blocking the interaction of CD40L with CD40 on B cells, thereby inhibiting cell proliferation and survival signals. It also induces antibody-dependent cell-mediated cytotoxicity (ADCC). Bendamustine crosslinks DNA, disrupting DNA/RNA synthesis and repair, leading to cell death via both apoptotic and nonapoptotic mechanisms. This combination is being investigated primarily for the treatment of **chronic lymphocytic leukemia (CLL)** and potentially other B-cell malignancies[5][2][4][6].
