LY-DMPC:DMPG-MSV is an experimental nanoparticle-based therapeutic designed for the treatment of pancreatic cancer. It consists of a TGF-β receptor Type 1 (TGF-β R1) inhibitor, identified as galunisertib (LY2157299), encapsulated within liposomes composed of dimyristoylphosphatidylcholine (DMPC) and dimyristoylphosphatidylglycerol (DMPG). These liposomes are further loaded into multistage silicon vectors (MSV), a drug delivery platform developed to provide sustained release and long-lasting inhibition of the TGF-β signaling pathway. In the pancreatic cancer tumor microenvironment, TGF-β signaling is a major driver of fibro-genesis and immune suppression, largely mediated by pancreatic stellate cells. By targeting TGF-β R1, LY-DMPC:DMPG-MSV aims to inhibit tumor cell proliferation, migration, and invasion while modulating the fibrotic stroma. Preclinical studies in murine xenograft models have demonstrated its efficacy in inhibiting the growth of pancreatic cancer tumors.
