LY2523355 (litronesib) is an allosteric inhibitor of Eg5 (kinesin spindle protein), a mitotic motor kinesin essential for bipolar spindle formation during cell division. It disrupts mitosis in rapidly dividing cancer cells by inhibiting Eg5 function. Pegfilgrastim is a long-acting recombinant human granulocyte colony-stimulating factor (G-CSF) analog that stimulates the production and maturation of neutrophils to reduce the risk of infection and febrile neutropenia associated with myelosuppressive chemotherapy. The combination is studied in advanced or metastatic cancers to allow higher dosing or mitigate neutropenia caused by LY2523355[1][3][4].
