LY2584702 + everolimus is an experimental combination therapy comprising LY2584702, an oral, selective ATP-competitive inhibitor of p70 S6 kinase, and everolimus, a well-established inhibitor of the mammalian target of rapamycin (mTOR). This combination targets the PI3K/AKT/mTOR signaling pathway at multiple nodes, aiming to suppress tumor cell proliferation and survival more effectively than inhibition at a single node. Preclinical data demonstrated synergistic antitumor activity for the combination, but phase I clinical study in patients with advanced solid tumors indicated minimal clinical benefit. LY2584702 was developed by Eli Lilly and Company; everolimus is also developed and marketed by Novartis, primarily as Afinitor for various indications, including solid tumors and certain types of cancer[1][3][5][7].
