LY2875358 + trametinib is an investigational combination therapy targeting cancer cell growth and survival pathways. LY2875358 is a neutralizing and internalizing monoclonal antibody directed against the c-MET receptor, developed to block hepatocyte growth factor (HGF)-mediated activation of c-MET signaling, which can drive tumor progression and resistance to other therapies. Trametinib is an orally available small molecule inhibitor of mitogen-activated protein kinase kinases 1 and 2 (MEK1/2), key components of the RAS-RAF-MEK-ERK pathway that regulates cell proliferation and survival. The combination has been studied preclinically for its ability to overcome HGF-mediated resistance to MEK inhibition in cancers such as uveal melanoma, with each agent targeting complementary mechanisms involved in tumor growth[6][8]. Trametinib is approved for use as monotherapy or in combination with dabrafenib for BRAF-mutant melanoma, non-small cell lung cancer, thyroid cancer, solid tumors, and certain brain tumors[1][3][5].
