LY3031207 + simvastatin is a combination therapy involving LY3031207, an investigational antisense oligonucleotide (ASO) targeting angiopoietin-like 3 (ANGPTL3) mRNA, and simvastatin, a standard-of-care HMG-CoA reductase inhibitor (statin). LY3031207 was developed by Eli Lilly to inhibit the hepatic production of ANGPTL3, a protein that regulates lipid metabolism by inhibiting lipoprotein lipase and endothelial lipase. By reducing ANGPTL3 levels, the drug aims to lower triglycerides and LDL cholesterol. Simvastatin acts by inhibiting the rate-limiting enzyme in cholesterol biosynthesis, thereby reducing endogenous cholesterol production. The combination was primarily evaluated in clinical trials to assess the safety, pharmacokinetics, and potential additive lipid-lowering effects in patients with hypercholesterolemia. Eli Lilly discontinued the development of LY3031207 in 2019.
