Overview

LY3200882 + capecitabine is an investigational combination therapy being evaluated for the treatment of advanced, chemotherapy-resistant colorectal cancer (CRC) in patients with an activated TGF-beta signature. This combination is specifically studied within Stratum 3 of the MoTriColor platform trial, led by the Netherlands Cancer Institute. LY3200882 is a small molecule inhibitor of the Transforming Growth Factor-beta (TGF-beta) receptor type I (ALK5), designed to block TGF-beta signaling, which often promotes tumor progression and immune evasion in CRC. Capecitabine is an oral fluoropyrimidine carbamate that functions as a prodrug of 5-fluorouracil (5-FU), an antimetabolite that inhibits thymidylate synthase and interferes with DNA synthesis. The rationale for the combination is to utilize TGF-beta inhibition to overcome resistance mechanisms and enhance the efficacy of cytotoxic chemotherapy in molecularly selected patients.
