Capecitabine is an oral fluoropyrimidine carbamate that functions as a prodrug of 5-fluorouracil (5-FU), an antimetabolite that inhibits thymidylate synthase and interferes with DNA synthesis. The rationale for the combination is to utilize TGF-beta inhibition to overcome resistance mechanisms and enhance the efficacy of cytotoxic chemotherapy in molecularly selected patients.
