Overview

LY3200882 + gemcitabine + nab-paclitaxel is a combination regimen investigated primarily in the treatment of advanced pancreatic cancer. LY3200882 is a selective, next-generation small molecule inhibitor of transforming growth factor beta receptor 1 (TGFβR1), acting as an ATP-competitive inhibitor of the serine-threonine kinase domain of the receptor, which interferes with TGFβ-mediated signaling known to play a significant role in tumor progression, metastasis, and immune suppression. Gemcitabine is a nucleoside analog used as an antimetabolite chemotherapy agent, inhibiting DNA synthesis. Nab-paclitaxel (albumin-bound paclitaxel) is a microtubule inhibitor that promotes tubulin assembly and disrupts mitosis. The combination was studied in a phase I clinical trial (NCT02937272) for newly diagnosed advanced pancreatic cancer, showing a 75% disease control rate, with LY3200882 believed to act through both direct antitumor and immunomodulatory mechanisms alongside standard cytotoxic chemotherapies[1][3].
