Overview

Combination of **LY3321367**, an anti-TIM-3 monoclonal antibody, and **LY3300054**, an anti-PD-L1 monoclonal antibody, both developed by Eli Lilly for use as an immuno-oncology therapy. LY3321367 blocks the TIM-3 immune checkpoint on T cells and other immune cells, aiming to reverse T cell exhaustion and enhance immune responses against tumors. LY3300054 targets PD-L1 on tumor cells and immune cells, releasing suppression of cytotoxic T cells. The combination is designed to overcome resistance seen with monotherapy by co-inhibiting distinct immune checkpoints. It has mainly been studied in advanced relapsed/refractory solid tumors, especially those with microsatellite instability-high (MSI-H) or mismatch repair-deficient (dMMR) profiles; the combination has shown a tolerable safety profile and some preliminary antitumor activity, particularly in PD-1/PD-L1 inhibitor-naïve populations[1][2][3][4][6].
