LY3325656 + sitagliptin is a combination therapy consisting of LY3325656 (a novel, investigational dual GIP and GLP-1 receptor agonist developed for the treatment of type 2 diabetes mellitus) and sitagliptin (a dipeptidyl peptidase 4 inhibitor that increases incretin hormone activity by preventing breakdown of GLP-1 and GIP). LY3325656 acts as a dual agonist for the receptors Glucose-dependent insulinotropic polypeptide (GIP) and Glucagon-like peptide 1 (GLP-1), improving glycemic control primarily by promoting insulin secretion and suppressing glucagon release. Sitagliptin augments this mechanism by inhibiting DPP-4, thus potentiating endogenous GLP-1 and GIP signaling. This combination is being investigated for synergistic effects in type 2 diabetes, aiming for enhanced glycemic control through complementary incretin pathway modulation. LY3325656 is developed by Eli Lilly, while sitagliptin is developed and manufactured by Merck.
