Overview

LY3410738 + esomeprazole is a combination therapy consisting of LY3410738, a potent and selective cholecystokinin-2 (CCK2) receptor antagonist (also known as the gastrin receptor), and esomeprazole, a proton pump inhibitor (PPI). The combination was developed by Eli Lilly to address limitations of PPI monotherapy in patients with acid-related disorders, such as gastroesophageal reflux disease (GERD). By blocking the CCK2 receptor, LY3410738 aims to inhibit the gastrin-mediated stimulation of gastric acid secretion and potentially mitigate the hypergastrinemia often observed during chronic PPI use. Clinical development focused on evaluating the synergistic effect of dual acid suppression mechanisms, particularly in patients who do not achieve adequate symptom relief with PPIs alone.
