LY3549492 is an investigational oral small molecule Glucagon-like peptide-1 receptor (GLP-1R) agonist being developed by Eli Lilly for the treatment of obesity and Type 2 Diabetes Mellitus, functioning by mimicking endogenous GLP-1 to enhance glycemic control and promote weight loss[1][3]. Atorvastatin is an oral small molecule HMG-CoA reductase inhibitor widely approved to lower cholesterol and reduce cardiovascular risk. Midazolam is an oral small molecule benzodiazepine acting as a GABA-A receptor positive allosteric modulator, used mainly as a probe drug in clinical pharmacology to assess drug-drug interactions or CYP3A activity. Combination use of LY3549492, atorvastatin, and midazolam often occurs in drug-drug interaction studies to evaluate the effects of LY3549492 on the pharmacokinetics of CYP3A4 and CYP450 substrate drugs (atorvastatin and midazolam), but this is not a marketed combination product[7].
