LY3556050 + metformin is an orally administered investigational combination comprising LY3556050, a selective and potent somatostatin receptor 4 (SSTR4) agonist, and metformin, a well-established biguanide used in diabetes. LY3556050 is under clinical development primarily as an analgesic for chronic pain conditions, particularly diabetic peripheral neuropathic pain (DPNP), osteoarthritis (OA), and chronic low back pain (CLBP). Several studies have evaluated the pharmacokinetic interaction and safety of the co-administration of LY3556050 and metformin, with findings that metformin exposure may be affected by LY3556050 coadministration and that metformin dosing may be limited in combined regimens due to drug-drug interaction concerns. The combination targets pain reduction through SSTR4 agonism with LY3556050 and glycemic control through metformin.
