LY6K + VEGFR1 + VEGFR2 is an investigational multiple peptide cancer vaccine developed by the Japanese Foundation for Cancer Research. It consists of three HLA-A*2402-restricted epitope peptides targeting lymphocyte antigen 6 complex locus K (LY6K), vascular endothelial growth factor receptor 1 (VEGFR1), and vascular endothelial growth factor receptor 2 (VEGFR2). LY6K is a cancer-testis antigen frequently overexpressed in esophageal squamous cell carcinoma (ESCC), while VEGFR1 and VEGFR2 are angiogenic receptors expressed on tumor-associated endothelial cells. The vaccine is designed to induce cytotoxic T lymphocyte (CTL) responses against both tumor cells and the tumor vasculature to overcome immune escape mechanisms like HLA downregulation. It is administered subcutaneously in combination with GM-CSF (sargramostim) as an adjuvant and is specifically restricted to patients who are HLA-A*2402-positive.
