Overview

Lysine-3,4-methylenedioxymethamphetamine (Lys-MDMA) is a small molecule prodrug of MDMA (3,4-methylenedioxymethamphetamine) developed by the University Hospital Basel and licensed to Mind Medicine. It was designed as a lysine-conjugated amphetamine intended to slowly release active MDMA upon absorption, thereby potentially enhancing tolerability, reducing abuse potential, and mitigating rapid physiological changes (such as blood pressure spikes) at the onset of drug action. While MDMA is a potent releaser of serotonin, norepinephrine, and dopamine being investigated for the treatment of post-traumatic stress disorder (PTSD), Lys-MDMA was found to be pharmacologically inactive in a Phase 1 crossover trial in healthy volunteers, as it failed to produce detectable MDMA plasma concentrations or clinical effects.
