Drug intelligence / Profile preview
**LYT-200 + venetoclax** is an investigational combination therapy for relapsed/refractory acute myeloid leukemia (AML) and high-risk myelodysplastic syndromes (MDS). **LYT-200** is a fully human IgG4 monoclonal antibody that binds to and antagonizes galectin-9, a protein implicated in promoting oncogenesis and inducing immune suppression in cancer. Downregulation of galectin-9 by LYT-200 exerts a dual mechanism of direct anti-leukemic effect (via induction of apoptosis and DNA damage) and reactivation of immune effector pathways, including NK and T cells. **Venetoclax** is a selective, small-molecule BCL-2 inhibitor that induces apoptosis in cancer cells dependent on BCL-2 for survival. This combination is being clinically evaluated in a phase 1 trial for relapsed/refractory AML and high-risk MDS, either with or without hypomethylating agents, aiming to improve outcomes in patients with poor prognosis and resistance to standard therapies[1][2][3][5]. Early clinical results indicate favorable safety and evidence of efficacy, including response and disease stabilization in patients previously refractory to standard treatments[1][2].
