M01ZH09 (Emergent BioSolutions)

M01ZH09 is a live attenuated oral vaccine candidate developed for the prevention of typhoid fever. It is derived from the *Salmonella enterica* serovar Typhi Ty2 strain and features two specific genetic deletions: the *aroC* gene, which renders the bacteria auxotrophic for aromatic amino acids, and the *ssaV* gene, a component of the Salmonella pathogenicity island 2 (SPI-2) type III secretion system. These modifications are designed to prevent systemic spread and render the strain replication-deficient while maintaining its ability to infect macrophages and induce a robust mucosal and systemic immune response. Originally developed by Microscience Ltd (later acquired by Emergent BioSolutions), the vaccine is administered as a single oral dose.
