Overview

The M1 positive allosteric modulator (including the lead compound ASN-408) is a small molecule drug being developed by Asceneuron to treat cognitive deficits in neurodegenerative conditions such as Parkinson's disease dementia (PDD), Lewy Body Dementia (LBD), and Frontotemporal dementia (FTD). It functions by binding to an allosteric site on the M1 muscarinic acetylcholine receptor (M1 mAChR), which enhances the receptor's affinity for its natural ligand, acetylcholine. This mechanism potentiates cholinergic signaling specifically at the M1 subtype—a target critical for memory and executive function—while avoiding the adverse side effects typically caused by non-selective activation of other muscarinic receptor subtypes (M2-M5). The compound has shown high specificity and nanomolar potency in cellular assays and neuronal cultures.
