Suven Life Sciences is developing a small molecule positive allosteric modulator (PAM) of the muscarinic M1 receptor for the treatment of gastrointestinal disorders. Currently in the lead optimization stage of preclinical development, this program aims to leverage the role of M1 receptors in regulating gastrointestinal motility and secretions. Unlike orthosteric agonists, which bind to the same site as the endogenous neurotransmitter acetylcholine and can lead to receptor desensitization or off-target effects, PAMs bind to a distinct allosteric site. This binding enhances the receptor's affinity or efficacy for acetylcholine, potentially providing a more physiological and selective therapeutic effect. This approach is intended to address various gastrointestinal conditions where cholinergic signaling is impaired, while potentially minimizing side effects associated with non-selective muscarinic activation.
